新德里金属β内酰胺酶-1氨基酸序列和基因序列的生物信息学分析
Bioinformatic Analysis of Amino Acid and Genetic Sequences of New Delhi metallo-β-lactamase-1
新德里金属-β-内酰胺酶-1(NDM-1)是"超级细菌"中抵抗常见抗生素的重要功能蛋白。了解NDM-1的功能基因来源及在不同物种间转移具有重要的生物学意义。本文旨在从氨基酸和基因序列分析NDM-1的可能进化模式。通过对一系列金属-β-内酰胺酶的进化树分析,发现YP_458405.1、YP_003270512.1和YP_003061440.1三个蛋白与NDM-1具有较近的氨基酸序列相似性,而YP_458405.1与NDM-1有较近的基因距离。进一步的GC含量分析也发现NDM-1上游100~350bp剧烈波动,而在编码区段与YP_458405.1的GC含量较为相似。因此,认为NDM-1基因可能来自YP_458405.1的水平转移。
New Delhi metallo-β-lactamase-1 (NDM-1) is the functional gene that is reponsible for drug-resistence in super bacteria. Understanding the genetic origin of the functional gene and possibility of gene transfer is highly critical in biology. This work is to explore the molecular evolution based on amino acid and genetic sequence of New Delhi metallo-β-lactamase-1 (NDM-1). Phylogenetic tree suggests that proteins YP_458405.1, YP_003270512.1, and YP_003061440 are relative of NDM-1, but only YP_458405.1 shares similar genetic coding with NDM-1. In addition, GC content analysis also shows that 100~350 bp upstream of NDM-1 gene fluctuates drastically, while the coding region is consistent with that of YP_458405.1. Thus, the study implied that NDM-1 may stem from horizontal transfer from YP_458405.1
