In order to study the regulation of sly-miR167a promoter, about 2000 bp fragment upstream of transcription start was amplified from tomato genomic DNA by PCR. It was fused to GUS and transformed into tomato (cv Microtom). The temporal and spatial specificity during plant development were analyzed. To investigate the response to exogenous hormone, treatments were performed on transgenic tomato. Promoter analysis revealed that some conserved regulatory sequence and several regulatory elements appeared in the promoter fragment. The results indicated that promoter miR167a was expressed in all the tissues, and expressed intensely in leaves, stems, flowers at anthesis and 2 days before anthesis, and fruits at immature green, mature green and breaker stages, and it was up-regulated by ethylene, NAA and GA. This result reveals that the expression of sly-miR167a promoter showed temporal and spatial specificity, and it is regulated by exogenous hormone.
