本文克隆了Klebsiella pneumoniae DSM2026的醛还原酶基因yqhD并在E. coli BL21(DE3)中得到了高效表达,经过Q-Sepharose和Sephacryl S-300两步柱层析纯化得到电泳条带单一的纯酶。活性检测表明该酶为醛还原酶,催化3-羟基丙醛等多种底物还原生成相应的醇。以3-羟基丙醛为底物,该酶的最适pH为5.8,最适温度为60 ℃,对NADPH和NADH的Km值分别为0.07 mM和1.42 mM。
In this paper Klebsiella pneumoniae yqhD was cloned and overexpressed in E. coli BL21(DE3). The enzyme was purified to homogeneity by Q-Sepharose and Sephacryl S-300. The catalytic characteristics of the enzyme were studied and it was an aldehyde reductase. Using 3-hydroxyaldehyde as substrate, the optimal pH and optimal temperature of the enzyme were pH 5.8 and 60 ℃, respectively, and the Km values for NADPH and NADH were 0.07 mM and 1.42 mM, respectively, which demonstrated the flexibility of dependence on coenzymes for this enzyme.
