目的 研究小檗碱(berberine,BBR)对肥胖小鼠脂肪组织中巨噬细胞(ATM)浸润的影响与胰岛素抵抗的关系。方法 高脂喂养肥胖C57BL/6小鼠分为模型组(DIO组)和BBR治疗组(DIO+BBR组),另设正常小鼠组(NC组)给予正常饮食。DIO+BBR组每日按10mgokg-1体重腹腔注射BBR,NC组和DIO组以等量的生理盐水腹腔注射,治疗2周。禁食过夜,取血清,分别检测血清血糖(FBG)和胰岛素(FINS)并计算胰岛素抵抗指数(HOMA-IR);另取附睾脂肪组织,用流式细胞术检测细胞表面的F4/80和CD11c的表达率。 结果 与DIO组比较,DIO+BBR组胰岛素抵抗指数(HOMA-IR)明显下降,FBG和FINS也有下降趋势,附睾脂肪组织间的F4/80+细胞和F4/80+CD11c+双阳性细胞均显著减少。同时与DIO组相比,BBR组小鼠血清MCP-1的表达显著降低。结论 BBR能够显著抑制巨噬细胞促炎因子MCP-1的分泌和巨噬细胞浸润,改善慢性炎症,进而改善胰岛素抵抗。

Objective: To observe the influence of berberine on infiltration of macrophages in adipose tissue of diet induced obesity mice. Methods: C57BL/6 mice were respectively fed with high fat diet and normal diet for 12 weeks. According to body weight, mice were divided randomly to DIO model group (DIO) and berberine treatment group (DIO+BBR). Normal mice fed with normal diet were set as control group (NC). Results: Compared with DIO group, berberine treatment significantly decreased the HOMA-IR factor, and slightly decreased FBG and FINS. At the same time, F4/80+ macrophages and F4/80+CD11c+ cells in epididymis adipose tissue were decreased significantly under berberine treatment compared with DIO group. Serum MCP-1 concentration in BBR group was decreased sharply compared with NC group. Conclusion:Berberine may decrease the secretion of MCP-1 of macrophages, inhibit macrophages infiltration into adipose tissue, then improve insulin resistance and glucose metabolism.
