三唑磷对斑马鱼miR-99及其预测靶标ckmt2的表达的影响
he impact of Triazophos on expression of miR-99 and predicted target gene ckmt2 in zebrafish
在农药三唑磷作用下,miRNA的表达发生改变,miRNA会进一步与靶基因碱基互补配对,调控基因的表达。采用荧光定量PCR等方法研究miR-99及其潜在靶基因ckmt2的表达变化。三唑磷处理后miR-99表达是其对照的0.062倍,助剂处理是其对照组的0.414倍,表达下调,三唑磷处理后ckmt2基因是其对照的0.91倍,助剂处理是其对照的0.89倍,在mRNA水平变化不显著。通过蛋白含量测定、Western blot分析及spss数据分析ckmt2处理前后的表达情况,结果表明三唑磷及助剂处理下,总蛋白含量减少,ckmt2编码的蛋白含量增加,采用独立样本检验方法分析结果表明在蛋白水平差异显著。综上所述,在三唑磷作用下,斑马鱼miR-99与其潜在靶标基因 ckmt2可能存在抑制作用,miR-99表达的减少使ckmt2受到的抑制减少,从而大量翻译。
he expression of miR-99 will change after treated with triazophos, and miRNA will regulate the gene expression through the base complementary matching. The expression change of miR-99 and its predicted target gene ckmt2 in zebrafish were evaluated using real-time PCR method after treated with triazophos. The results showed that the expression of miR-99 was declined, which is 0.062 times and 0.414 times the control for triazophos and adjuvant group.Ckmt2 gene expression did not change significantly, which is 0.91 and 0.89 times the control for triazophos and adjuvant group. However, the protein concentration measurement, Western blot and SPSS analysis indicated that the total protein content is decreased and protein coded by ckmt2 is increase after treated with triazophos. the result is significant by analyzed using independent sample test analyzed.In summary, ckmt2 gene expression may be inhibited by miR-99 in zebrafish, in function of triazophos, miR-99 expression increased, and the inhibition of ckmt2 is reduced, and then resulting in a heavy translation.
