腺病毒技术封闭smad4基因表达对人原代血管内皮细胞增殖和迁移的影响
Influence of adenovirus-induced smad4 gene silencing on proliferation and migration of HVEC cell
背景与目的 探讨利用腺病毒技术下调smad4基因表达对人原代血管内皮细胞增殖和迁移能力的影响,初步了解smad4在血管生成中的作用。方法 设计并合成靶向人smad4基因的腺病毒Adv-smad4,转染入人原代血管内皮细胞HVEC中,荧光显微镜检测转染效率,Western Blot检测转染前后smad4基因表达的水平; CFSE-流式细胞仪检测ECV304 细胞转染前后细胞增殖能力的变化;单层伤口愈合实验观察细胞转染前后迁移能力的改变。结果 Adv-smad4能够明显下调smad4基因表达,HVEC细胞转染Adv-smad4后,细胞的增殖和迁移能力明显增强。结论 利用腺病毒技术能够高效特异阻断smad4基因表达,smad4基因表达下调能够明显促进血管内皮细胞的增殖和迁移。
Background and Objective:To investigate the effect of silencing smad4 gene expression by using adenovirus on proliferation and migration of HVEC cell. Methods : Adenovirus targeting smad4 gene were designed. Adv-smad4 was transfected into HVEC cells. The alteration of smad4 expression was examined by Western blot. The proliferation capacity of HVEC cells was measured by CFSE-flow cytometry. The monostratal wounding healing was used to determine the migration capacity of HVEC cells. Results: Adv-smad4 was successfully transfected into HVEC cells. Transfection of Adv-smad4 significantly knocked down the smad4 protein expression. The proliferation and migration capacity of HVEC cells transfected with Adv-smad4 was significantly enhanced compared with that of control group. Conclusions:The specific smad4-targeted adenovirus can efficiently reduce endogenous smad4 gene expression in HVEC cells. Selectively smad4 gene silencing can promote the proliferation and migration of HVEC cells.
