Study on Damage Effects of Mitochondria of Cardiomyocytes Induced by Cadium

目的:研究CdCl2对心肌细胞线粒体的损失作用。方法:原代培养大鼠心肌细胞,用MTT检测细胞存活率,生物化学方法检测ATP酶和LDH的活性;流式细胞术检测膜电位的变化,分光光度法测量膜通透性的改变。结果:5µmol/L CdCl2 即可使心肌细胞存活率降低,ATP酶和LDH活性降低,并且膜电位和膜通透性都发生改变。所有剂量组各指标和对照组相比较差异具有显著性(P<0.05)。结论:CdCl2 可诱导心肌线粒体损失,并且可能存在剂量依赖关系。

Objective:To study damage effects of mitochondria of cardiomyocytes induced by CdCl2. Methods:Incubate cardiomyocytes of rats with the primary cell culture method. Examine survival rate of myocytes with MTT assay, and detect avtivities of ATP enzyme and LDH with biochemical method, MMP with FCM, MPT with spectrophotometry. Results: In exposure groups, 5µmol/L CdCl2 could decrease survival rate of cardiomyocytes, activities of ATP enzyme and LDH, membrane potential of mitochondria, and membrane permeability of mitochondria was changed induced by CdCl2. There were statistically significant differences between control and exposure groups (P<0.05). Conclusions: CdCl2 could induce damage of mitochondria of cardiomyocytes, and there possibly exits a dose dependent relationship.
