miR-126在高糖诱导的脉络膜视网膜内皮细胞应激中的作用研究

目的:miR-126作为内皮细胞和造血祖细胞特异性表达的miRNA,与血管生成密切相关。本研究旨在研究miR-126在高糖诱导的脉络膜视网膜内皮细胞应激中的作用机制。方法:以30 mM高糖培养液培养脉络膜视网膜内皮细胞(RF/6A)模拟高糖环境,链脲佐菌素诱导建立糖尿病大鼠模型。用Real time-PCR检测细胞和视网膜组织中miR-126表达。利用Lipofectamine2000试剂和OptiMEM I低血清培养液将miR-126-mimic转染RF/6A细胞。应用免疫印迹法检测miR-126下游靶基因VEGF蛋白的表达。结果:高糖培养可明显诱导RF/6A细胞miR-126表达下调。糖尿病大鼠视网膜组织中miR-126表达较非糖尿病大鼠明显降低。高糖培养诱导RF/6A细胞VEGF表达增加,而上调miR-126可缓解高糖诱导的RF/6A细胞VEGF表达增加。结论:本研究发现在高氧环境下miR-126表达降低,上调miR-126可通过抑制高糖诱导的VEGF表达减缓新生血管形成,证实了miR-126在糖尿病视网膜病变视网膜新生血管过程中的重要临床价值。
Objective: miR-126, the miRNA considered to be specially expressed in endothelial cells and hematopoietic progenitor cells, is strongly associated with angiogenesis. The purpose is to evaluate the role of miR-126 in high glucose-induced angiogenesis and the possible mechanisms. Methods: The expression of miR-126 was detected in high glucose-treated RF/6A cells and diabetic retinas using real-time PCR. The miR-126 was up-regulated by transfecting miR-126-mimics into RF/6A cells. The protein level of vascular endothelial growth factor (VEGF) was assessed by immunoblotting. Results: A significantly decreased expression of miR-126 was found in high glucose-treated RF/6A cells compared with control. The expression of miR-126 was also reduced in the retina tissue of streptozotocin-induced diabetic rats. The expression of VEGF protein was increased in high glucose-induced RF/6A cells. The VEGF protein level was sharply decreased in high glucose-induced RF/6A cells pretreated with miR-126-mimics. Conclusions: miR-126 is down-regulated under high glucose condition both in vitro and in vivo and may halt the high glucose-induce neovascularization by inhibiting the expression of VEGF.
