筛选家蚕蛹cDNA文库,得到一条脂蛋白的cDNA序列,其命名为BmLp-c21(Bombyx mori lipoprotein pBmHPC-21)。该基因全长为2803 bp,ORF框为792 bp,编码263个氨基酸残基,预测该蛋白的分子量为30.241 kD,等电点为7.23。我们将BmLp-c21基因克隆至原核表达载体pET-28a(+),转化大肠杆菌Rosetta(DE3),IPTG诱导表达融合蛋白,通过亲和层析得到纯化的融合蛋白,用该纯化的融合蛋白免疫新西兰大白兔制备多克隆抗体,ELISA检测效价可达1:6400。亚细胞定位显示,BmLp-c21主要存在于细胞质中。通过Western blotting 和实时荧光定量PCR的方法,发现BmLp-c21在家蚕不同发育阶段和五龄幼虫不同组织中的转录和表达水平存在较大差异。BmLp-c21在家蚕蛹期及五龄幼虫血液中的转录和表达水平最高。本研究为进一步探讨BmLp-c21基因在家蚕生长、发育过程中的生物学功能打下了一定的基础。

cDNA sequence which encodes lipoprotein was found from the pupa cDNA library, the gene was named BmLp-c21 (Bombyx mori lipoprotein pBmHPC-21). The length of the cDNA is 2803 bp, the open reading frame(ORF) is 792 bp and encodes a protein of 263 amino acid residues with the predicted molecular weight of 30.241 kD and isoelectric point of 7.23. BmLp-c21 was inserted into the expression vector pET-28a (+) to construct the recombinant plasmid. The fusion protein was expressed in the E.coli Rosetta (DE3) induced by IPTG, and then was purified by affinity chromatograph. Polyclonal antibody was generated by immunizating New Zealand rabbit with the purified fusion protein .The titer of the polyclonal antibody was larger than 1:6400, measured by ELISA. Immunofluorescence method was employed to detect the subcellular localization of BmLp-c21 in BmN cells. We found that BmLp-c21 was distributed mainly in the cytoplasm. We found that the transcription and expression level of BmLp-c21 were different in the different tissues of the fifth instar larva and developmental stages of Bombyx mori by real-time PCR and Western blotting. The result revealed that: BmLp-c21 gene transcripts and expresses higher in pupa than other stages and higher in blood compared to other tissues of the fifth instar larva. It laid a foundation for further Studies on BmLp-c21 gene.
