酸刺激对食管腺癌相关基因NOX5-S表达的影响研究

目的 研究酸刺激下食管腺癌相关基因NOX5-S的表达,了解食管腺癌的发病机制。方法 1、将培养的OE33食管腺癌细胞株随机分为2组:实验1组(PH=4.0)和实验2组(PH=4.0,同时去除培养基中Ca2+),分别测量并比较两组间的活性氧簇、NOX5-S mRNA及蛋白的表达水平。2、将培养的OE33食管腺癌细胞株随机分为2组:实验1组(PH=7.2)和实验2组(PH=4.0),分别测量并比较两组CREB mRNA及蛋白水平的表达水平。结果 1、实验1组的NOX5-S和ROS水平明显比实验2组高(P<0.05);2、实验2组CREB表达水平明显比实验1组高(P<0.05)。结论 酸刺激OE33食管腺癌细胞引起细胞内的Ca2+浓度升高,CREB及NOX5-S的表达增加。
Objective To research the expression of NOX5-S in esophagus adenocarcinoma cells and understand the mechanisms of esophagus adenocarcinoma. Methods 1.The cultured OE33 esophageal adenocarcinoma cell lines were randomly divided into 2 groups: GroupⅠ(PH=4.0) and Group Ⅱ(PH=4.0,deplete of Ca2+ in the mediun), then measured and compared the level of NOX5-S mRNA and Protein and ROS between the two groups. 2. The cultured OE33 esophageal adenocarcinoma cell lines were randomly divided into 2 groups:GroupⅠ(PH=7.2) and Group Ⅱ(PH=4.0),then measured and compared the level of CREB mRNA and Protein between the two groups. Results 1.The level of NOX5-S and ROS in GroupⅠwas significantly higher than Group Ⅱ(P<0.05). 2.The level of CREB in Group Ⅱwas significantly higher than GroupⅠ(P<0.05). Conclusion Acid treatment could increase the density of intracellular Ca2+ in OE33 esophageal adenocarcinoma cell, the expression of CREB and NOX5-S was increased.
