高效液相色谱法测定茶条槭悬浮细胞中没食子酸含量

茶条槭悬浮培养细胞经过10%浓硫酸-甲醇提取后,用HPLC法测定其没食子酸含量。色谱条件为:流动相为甲醇/水(4/6),流速0.5ml/min,Waters C18 MS柱,检测波长为270nm。实验测得的线性范围为 20.0-100μg•mL-1(R=0.9996, n=5)。平均加样回收率为99.86%,RSD为0.09%。样品稳定性、实验的精密度及重复性三者的RSD均小于1.5%。表明本法是一种测定茶条槭悬浮细胞中没食子酸含量的好方法。

HPLC procedure was established to determin the content of gallic acid in suspension cells of Acer ginnala. The separation was performed on Waters - C18 column with a mobile phase of methanol/water (4/6) and detected at wavelength of 270 nm. The results showed that the linearity range was 20.0-1000μg•mL-1 (R=0.9996, n =5) and the average recovery of gallic acid was 99.86% , RSD was 0.09%. RSD values of sample stablity, precision and repetition were below 1.5%. All these results indicated that this preocedure works well.
