同型半胱氨酸作用下的人脐静脉内皮细胞凋亡相关基因表达谱研究

目的 从基因组水平对同型半胱氨酸(Homocysteine, Hcy)诱导血管内皮细胞凋亡的分子机制进行较深入和系统的研究。材料与方法 运用基因芯片技术对Hcy作用下的人脐静脉内皮细胞中凋亡相关基因表达谱进行了研究采。结果 获得了Hcy作用后凋亡相关基因表达谱,并检出表达下调基因12个,表达上调基因16个。结论 本次实验结果提示,Hcy诱导的细胞凋亡过程中可能主要涉及TNF受体通路分子,其中,主要包括Fas/CD85、TRAIL、TNFR1和DcR1(TRAIL受体)的表达上调。而在线粒体通路中,虽然Apaf-1表达升高,但caspase-9表达下降,并且细胞色素c释放减少,提示线粒体通路可能不是Hcy的主要信号通路。上述结论尚需进一步实验数据的支持。

bstract To understand the molecular mechanisms of the apoptosis of Vein endothelial cells caused by homocysteine, we use the apoptosis genechips to identify the gene expression profile of HUVEC pre-treated with homocysteine in-vitro. Based on the analysis of gene expression profile, we observed 16 up-regulated genes and 12 down-regulated genes in HUVEC incubated with Hcy. The results suggested that Fas/CD85、TRAIL、TNFR1 and DcR1 , which belong to the TNF family members, are increasing in HUVEC treated with HCY, this result suggested that TNF pathway may be play a big role in the process of apoptosis induced by Hcy. But the expression level of Apaf-1 is increasing , while caspase 9 expression is decreased and the release of cytochrome C from motochondrial is decrease, so mitochondrial pathway may be not a main pathay in apoptosis induced by Hcy.
