麻辣提取物对肠道微生物的影响

本实验探讨川菜的精髓麻辣提取物对小鼠肠道微生物的影响,分析小鼠灌喂麻辣提取物后肠道微生物的变化规律。研究将小鼠分成4 组,生理盐水对照组(CON)、水溶性提取物(HCAE)灌喂组、脂溶性提取物(HCFSE)灌喂组、粉末(HCP)灌胃组,连续灌喂35天。采集不同时间点小鼠粪便,利用高通量Illumina Miseq平台测序,对不同组别的小鼠肠道菌群作宏基因组分析。试验共获得18个小鼠粪便样本的93635个OTUs共1461778条读数。生物学分析结果表明,灌胃周期和灌胃物质不同的情景下,小鼠肠道细菌群落多样性存在差异, 且水溶性麻辣提取物对小鼠肠道菌群Alpha多样性的影响最大。在灌胃周期不同的情景下,初步推测小鼠肠道微生物对麻辣提取物有一个从抵触到逐渐适应的过程。表现在整个试验周期内,占主导地位的厚壁菌门与拟杆菌门的比例先减小,随后趋向平衡。但与同一时间的CON相比,HCP组、HCAE组以及HCFSE组的厚壁菌门、拟杆菌门的绝对条数都减少。
In this paper,the essence of Sichuan Cuisine extract on the intestinal microflora of mice wsa investigated, analyzing the variation of mouse intestinal microorganisms after gavaging spicy and hot ash extract. Research will be divided into four groups of mice, normal saline control group(CON), aqueous extract feed group(HCAE), fat-soluble extract feed group(HCFSE), powder lavage group(HCP), continuously fed with 35 day.Mice feces collected at different time points, using the second generation sequencing machine sequencing for different groups of mice intestinal macro genome analysis. Tested mice received 18 stool samples of 93635 OTUs,1461778 readings. Biological analysis results show that under the situation of differert lavage cycle and material , there are differences in mouse intestinal bacterial abundance and diversity of bacterial communities,and water soluble spicy and hot extract has the greatest impact on Alpha diversity of intestinal microflora in mice. In different contexts gavage period, initially speculated that mouse intestinal microbial have the process from resistance to gradually adapt to spicy and hot ash extract. This is manifested in the ratio of Firmicutes and Bacteroides first decreased and then tends to balance throughout the test period. However, compared with the absolute number of CON group at the same time, the number of Firmicutes and Bacteroides in other groups are all reduced .
