Study on photosynthetic characteristics of yellowing mutant in Brassica napus

本文调查分析了甲基磺酸乙酯EMS(ethyl methanesulfonate)诱变的甘蓝型油菜叶绿素含量减少黄化突变体crm1(chlorophyll-reduced mutant)的生育期、动态株高、叶片解剖特征、光合特性、叶绿素荧光动力学参数、叶绿素生物合成中间代谢产物含量等。结果表明,突变体crm1花期推迟约3天、生育期延长约4天,整个生育期株高变矮,株高为野生型的85.5%。crm1平展叶的显微结构不规则,栅栏组织和海绵组织松散,叶肉细胞间隙变大。crm1的光合速率、气孔导度和蒸腾速率都低于正常叶。叶绿素荧光动力学参数分析显示,crm1的F0、Fm、F0\'、Fm\'均极显著低于正常叶,而NPQ却显著提高,说明突变体PSII反应中心具有较高的热散耗。crm1叶绿素合成中间代谢产物Coprogen III升高,Proto-IX、Mg-Proto IX和Pchlide则降低,表明叶绿素合成受阻于Coprogen III到Proto IX这一步。

his paper investigated and analyzed the growth period, dynamic plant height, leaf anatomical characteristics, photosynthetic characteristics, chlorophyll fluorescence kinetic parameters, and chlorophyll biosynthesis intermediate metabolites content of the yellowing mutant crm1 (chlorophyll-reduced mutant) of Brassica napus induced by ethyl methanesulfonate EMS. The results showed that the flowering period of the mutant crm1 was delayed by about 3 days and the growth period was extended by about 4 days. The plant height of the mutant crm1 became shorter during the whole growth period, and the plant height was 85.5% of that of the wild type. The microstructure of crm1 flat leaf is irregular, palisade tissue and spongy tissue are loose, and mesophyll cell gap becomes larger. The photosynthetic rate, stomatal conductance and transpiration rate of crm1 were lower than those of normal leaves. The analysis of chlorophyll fluorescence kinetic parameters showed that F0, Fm, F0 \'and Fm\' of crm1 were significantly lower than those of normal leaves, while NPQ was significantly increased, indicating that the mutant PSII reaction center had a high heat dissipation. Crm1 Coprogen III, the intermediate metabolite of chlorophyll synthesis, increased, while Proto-IX, Mg-Proto IX and Pchlide decreased, indicating that chlorophyll synthesis was blocked by the step from Coprogen III to Proto IX.
