肾炎防衰液对UUO大鼠肾组织TGF-β1 和Smad7表达的影响

Influence of Shenyanfangshuai Solution on the expression of TGF-β1 and Smad7 in the obstructed kidney after unilateral ureteral obstruction (UUO )in rats

目的:动态观察肾炎防衰液对单侧输尿管梗阻(UUO) 所致肾间质纤维化大鼠转化生长因子β1(TGF-β1)和Smad7表达的影响。方法:111只SD大鼠随机分为正常组、假手术组、模型组、对照组、肾炎防衰液大、中、小剂量组。分别在术后第二周、第四周处死动物, 处死动物,观察肾脏病理改变。采用免疫组化方法检测肾组织中TGF-β1、Smad7表达。结果:与正常组相比,各时间点的模型组大鼠肾脏病理损害进行性加重, 肾炎防衰液治疗后可明显减轻模型组大鼠的肾肾间质纤维化程度(P<0.05)。随着梗阻时间的延长,模型组大鼠肾组织TGF-β1 表达增加;Smad7表达下降,与模型组各对应时间点比较, 肾炎防衰液治疗后可使肾组织中的Smad7表达显著增高,TGF-β1表达明显降低(P<0.05) 。结论:肾炎防衰液可能通过诱导Smad7表达上调及抑制TGF-β1表达,减轻肾间质纤维化。

Methods:The model rats were induced by ligating the left ureter. Male Sprague-Dawley ( SD )rats were divided into 7groups:the normal group,sham-operation group,model group,benazepril group,and three Xiaozhengsanjie zufang (Shenyanfangshuai Solution) groups(in different doses).Rats were sacrificed on the second and four week after UUO was initiated. Immunohistoehemistry method was performed to examine the expression of TGF-β1 in the obstructed kidney. Results:The expression of TGF-β1 increased but Smad7 decreased in the model group with the extension of obstruction; Compared twith the UUO group , the expression of TGF-β1 were decreased but Smad7 increased in the Shenyanfangshuai Solution groups , Conclusion:Shenyanfangshuai Solution significantly alleviates renal interstitial fibrosis by suppressing the expression of TGF-β1 and improving the expression of Smad7.
