金刚健骨片调节整合素β1、αⅤβ3表达水平的实验研究
Effect of Jingang Jiangu Pill on Levels of Intigrin β1 and αⅤβ3 Expression in Ovariectomized Osteoporosis Model Rats
目的:通过对去势大鼠整合素表达水平的观察,揭示金刚健骨片治疗骨质疏松症的作用机理。方法:将50只10月龄Wistar雌性大鼠随机分为5组:福善美组(FSM)、金刚健骨片组(JGJG)、骨松宝颗粒组(GSB)、模型组(OVX)、假手术组(SHAM)。治疗组及模型组行卵巢摘除术,假手术组切除相同重量的肠系膜。统一饲养13周造模成功后,治疗组分别以相应药物连续给药13周;模型组及假手术组以生理盐水替代。治疗结束后处死动物取出骨组织,进行骨密度测定、并利用免疫组化法测定整合素αⅤβ3及β1表达水平。结果:骨密度测定结果显示,治疗组的股骨密度、整合素β1均较OVX组有明显提高,差异具有统计学意义(P<0.05);治疗组整合素αⅤβ3明显低于OVX组,差异具有统计学意义(P<0.05)。结论:金刚健骨片能显著提高去势大鼠的股骨密度、整合素β1的表达水平,并可降低整合素αⅤβ3的阳性表达率,从而干预成骨-破骨细胞偶联,达到治疗目的。
Objective: To investigate the regulatory effect of Jingang Jiangu Pill(JGJG) on integrin of ovariectomized rats. Methods: 50 ovariectomized 10 months old female rats were randomly divided into 5 groups as Fushanmei group (FSM), Jingang Jiangu Pill group(JGJG), Gu Songbao Granule group(GSB), Model group(OVX), Sham group. After ovariectomized, the rats were raised in the same environment for 13 weeks. Then they were treated with different medicine for another 13 weeks. Bone mineral density(BMD) and the expression of integrin β1 and αⅤβ3 were detected in each group after the treatment.Results:BMD, the expression of integrin β1 of FSM group, JGJG group and GSB group were improved obviously than that of OVX group. There was statistical difference between these groups(P<0.05). The expression of integrin αⅤβ3 of the three treating group were significantly depressed. Conclusion: JGJG pill improves BMD and integrin β1 of ovariectomy rats and reduces their integrin αⅤβ3 through the regulation of the coupling of osteoblasts and osteoclasts.
