卡介菌多糖核酸(BCG-PSN)对小鼠的一般毒性和致突变研究

为探讨卡介菌多糖核酸(BCG-PSN)对应用动物的毒理学效应及其程度,本试验就BCG-PSN对ICR小鼠的急性毒性试验、亚慢性毒性试验和致突变作用进行了研究。结果表明,BCG-PSN小鼠腹腔注射的LD50 > 2000 mgokg-1obw-1,在其最大溶解度范围内无急性毒性;BCG-PSN对小鼠的亚慢性毒性试验表明,高剂量BCG-PSN组(1000 mgokg-1obw-1)小鼠在用药后的48 h内表现扎堆、畏寒、不喜运动、被毛蓬松等,以后逐渐好转的毒性反应;长期大剂量应用BCG-PSN会诱使动物出现轻微贫血和肾脏损伤,但在中、低剂量条件下长期使用是安全的;小鼠骨髓微核试验和精子畸形试验结果均呈阴性,表明BCG-PSN的致突变作用较弱。
o investigate the toxicological effect of Bacillus Calmette-Guerin -polysaccharide-nucleic acid (BCG-PSN) on animals, the acute toxicity, inferior-chromice toxicity, mutagenic actions of BCG-PSN to ICR mice were studied in this paper. The results revealed that the LD50 of BCG-PSN in mice is higher than 2000 mgokg-1obw-1 which is the maximum dissolubility of BCG-PSN, showing that there is no acute toxicity to mice. The inferior-chronic toxicity of high dose group(1000 mgokg-1obw-1)in mice revealed that BCG-PSN result in the toxic responses characterized by chilly, move reduction, the fur fluffiness, diarrhea and more eye secretion within 48 h after BCG-PSN injection. The slight anemia and weak kidney injure can be induced with high dosage and long time exposure to BCG-PSN injection, however there were no toxic responses with middle and low dose after long time exposure to BCG-PSN. The negative results of the cellular medullares micronucleus test and the teratospermia test demonstrated that BCG-PSN has low mutagenic action.
