玻璃体视网膜增殖膜中survivin 基因的表达研究
Expression of Survivin in the Preretinal Membranes
目的:探讨凋亡抑制基因survivin 在增生性糖尿病视网膜病变(PDR),增生性玻璃体视网膜病变(PVR),特发性黄斑视网膜前膜(IMPRM)增生膜中的表达.方法:玻璃体手术中取出的11例PDR,10例PVR,12例IMPRM患者的增生膜, 其中6例PDR,5例PVR,6例IMPRM患者的增生膜应用RT-PCR的方法检测增生膜中survivin 基因mRNA水平的表达。5例PDR,5例PVR,6例IMPRM患者的增生膜应用免疫荧光的方法检测增生膜中survivin 基因蛋白水平的表达以及定位。结果:6例PDR(100%),5例PVR(100%),6例IMPRM(100%)患者的增生膜中, survivin 基因mRNA水平均呈阳性表达。免疫荧光检测结果显示5例PDR(100%)患者的增生膜中survivin表达阳性,在血管内皮细胞(CD31),视网膜色素上皮细胞(cytokeratin PAN),胶质细胞(GFAP),成纤维细胞(vimentin)均表达阳性,阳性部位位于细胞核内,PDR增生膜中主要的细胞外基质fibronectin(FN), laminin(LN)与survivin无共表达。5例PVR(100%)患者的增生膜中survivin表达阳性,胶质细胞,成纤维细胞均表达阳性,阳性部位位于细胞核内。6例IMPRM(100%)患者的增生膜中survivin表达阳性,胶质细胞,成纤维细胞均表达阳性,阳性部位位于细胞核内。结论:: Survivin 基因的异常表达引起细胞的细胞增殖和凋亡抑制,在PDR,PVR和IMPRM 的发生发展中起一定作用,为临床PDR,PVR和IMPRM的治疗提供了新的靶点。
PURPOSE Proliferative diabetic retinopathy (PDR), Proliferative vitreoretinopathy(PVR) and idiopathic macular preretinal membrane(IMPRM) results from the formation of preretinal membranes (PRMs) in the posterior fundus that can lead to a severe decrease of vision. Survivin (SVV) is a protein that is highly expressed in the cells of tumors, but whether it plays a role in PRMs is unknown. The purpose of this study was to determine whether SVV is associated with the formation of PRMs. METHODS PRMs were obtained during vitrectomy from patients with PDR, PVR and IMPRM. RT-PCR was performed to determine the level of expression of the mRNA of SVV. Immunofluorescence analyses were performed to determine the sites of SVV in the PRMs. RESULTS The level of the mRNA of SVV was high in 6 of 6 PRMs from patients with PDR, 5 of 5 PRMs from patients with PVR and 6 of 6 PRMs from patients with IMPRM. Immunofluorescence analysis showed the colocalizations of SVV and CD31, an endothelial cell marker, SVV and cytokeratin PAN, a retinal pigment epithelial cell marker, SVV and Glial Fibrillary Acidic Protein(GFAP), a gial cell marker, SVV and vimentin, a fibroblast cell marker, SVV and fibronectin(FN), laminin(LN), two members of the extracellular matrix proteins in most of the cells in the PRMs from PDR. Immunofluorescence analysis showed the colocalizations of SVV and GFAP, SVV and vimentin, SVV and fibronectin, laminin in most of the cells in the PRMs from PVR. Immunofluorescence analysis showed the colocalizations of SVV and GFAP, SVV and vimentin in most of the cells in the PRMs from IMPRM. CONCLUSIONS This study indicates that SVV may play a significant role in the proliferation and maintenance of most cells in the PRMs. If correct, SVV may be a molecular target for new diagnostic and therapeutic strategies for PDR, PVR and IMPRM.
