壳寡糖进入细胞的机制研究

壳寡糖具有多种生物活性,如抗感染;促使癌细胞凋亡;调节免疫等。但是壳寡糖进入细胞的机制尚不清楚,现将壳寡糖进行荧光标记,观察壳寡糖进入细胞的微观机制。实验结果表明壳寡糖进入细胞受温度和细胞膜流动性的影响,升高温度使得壳寡糖进入细胞的含量增加,采用A23187加速细胞膜的流动可以明显地促进壳寡糖进入细胞内。另外,本文对A23187增加细胞膜磷脂酰丝氨酸外翻促进细胞膜流动进行了实验证明。

Oligosaccharides has varieties of biological activities, such as anti-infective; induce apoptosis in cancer cells; regulate immunity. But the mechanism of oligochitosan enter the cells is unclear, we oligochitosan fluorescently labeled observe the microscopic mechanism oligochitosan into cells. Experimental results show that chitosan oligosaccharide into the cell caused by membrane fluidity and temperature , when elevated temperatures makes oligochitosan enter the cell content increased, A23187 can significantly accelerate the oligochitosan into the cell by increase membrane fluidity. In addition, we prove that A23187 can facilitate membrane fluidity due to an increase in the plasma membrane phosphatidylserine concentration.
