为研究热冲击及其与铜联合对鲤鱼血清HSP70诱导影响,以健康1龄鲤鱼为受试生物,采用双抗体夹心酶联免疫吸附剂测定(Enzyme-Linked Immunosorbnent Assay ELISA)检测鲤鱼血清HSP70合成水平;原子吸收光谱法(Atomic Absorption Spectroscopy AAS)检测鲤鱼血清中铜残留量。试验结果表明,① 缓慢热冲击,鲤鱼血清HSP70诱导水平随温度的升高而增高;陡然热冲击,20℃组、24℃组血清HSP70水平与10℃空白对照组HSP70水平差异显著(p﹤0.01),28℃组HSP70水平与10℃空白对照组HSP70水平无显著性差异(p﹥0.05)。② 20℃热冲击和铜对HSP70合成无联合效应。

he present study aimed at determining the effects of heat shock and the combined effects of heat shock and copper on carp (Cyprinus carpio) serum. There are two parts in this paper. The first part consists of heat shock experiments, gradually heating and suddenly heating. In the second part, carp were exposed to different temperature levels (10℃、20℃、24℃) and six copper concentations (0、10、50、100、300、500ug/l). HSP70 expression was analyzed by sandwish ELISA (Enzyme-Linked Immunosorbnent Assay ELISA);Copper concentrations in carp serum were measured by atomic absorption spectroscopy (AAS). The results showed that, in the gradually heating experiment, serum HSP70 expression increased gradually as the temperature went up; in the suddenly heating experiment , the HSP70 expression in carp serum under 20℃and 24℃were both much higher than that under 10℃(P<0.01). There was no combined effects of 20℃ heak shock and copper on HSP70 expression.With the increase of copper dose , the expression of HSP70 did not change much compared to that of control group.(P>0.05).
