目的 研究黄药子及黄药子配伍当归后对大鼠肝CYP1A2、CYP2E1蛋白表达的影响。方法 给予成年SD大鼠黄药子及黄药子配伍当归后的药剂,观察肝组织病理形态的变化及以免疫组化法检测肝CYP1A2、CYP2E1的表达。结果 与空白对照组相比,黄药子组在短期内对大鼠造成肝损伤的同时,CYP1A2、CYP2E1的蛋白表达均显著升高。当黄药子配伍当归后,与黄药子组相比,肝损伤程度明显减轻, CYP1A2、CYP2E1蛋白表达显著降低。结论 在黄药子造成的肝损伤初期,强烈诱导了CYP1A2、CYP2E1的蛋白表达。而在黄药子配伍当归后,使激增的CYP1A2、CYP2E1的表达得以缓解,提示当归是通过抑制CYP1A2、CYP2E1的表达,对黄药子起到减毒抑毒作用的。

Objective To investigate the effects of Dioscorea bulbifera L. and medicine in group of Chinese angelica to Dioscorea bulbifera L. on the degree of the proteinic expressions of CYP1A2、CYP2E1 in rats liver.Methods The pathology results and proteinic expressions of CYP1A2 and CYP2E1 by the method of immunohistochemistry were observed in SD rats liver after the administration of Dioscorea bulbifera L. and medicine in group of Chinese angelica to Dioscorea bulbifera L. Results In the initial stages of the hepatic damnification aros的 Dioscorea bulbifera L.,it intensively induced the proteinic expressions of CYP1A2 and CYP2E1. The proteinic expressions of CYP1A2 and CYP2E1 were significantly lower in Chinese angelica to Dioscorea bulbifera L.group,and the hepatic damnification was obviously mitigated. Conclusion Chinese angelica lightens the hepatic toxicity by means of controlling the proteinic expressions of CYP1A2 and CYP2E1.
