为研究急性试验中三聚氰胺单独或与三聚氰酸协同对雄性小鼠精子质量的影响,本试验将42只7周龄SPF雄性昆明小鼠随机分为4组,三聚氰胺(MA)和三聚氰酸(CA)按1:1比例混合染毒,分别给予低剂量(30mg/kg)、中剂量(140mg/kg)和高剂量(700mg/kg),连续灌胃3d,玉米油作溶剂,对照组灌胃等量的溶剂玉米油。最后一次灌胃后24h,眼静脉采血,无痛处死小鼠。制备精子悬液,计算精子数量,活精子率,精子畸形率,单细胞凝胶电泳检测精子DNA损伤。结果显示,高剂量的MA-CA可以引起附睾精子数量显著下降;MA和MA-CA各剂量组均显著降低活精子率,显著增加精子畸形率,尤其是头部畸形率,并均表现出剂量效应;高剂量的MA及MA-CA引起精子DNA损伤显著。上述结果表明急性试验中MA与MA-CA均对雄性小鼠精子质量产生明显的损伤,且MA-CA对其损伤更为严重。

In order to study the effect of melamine alone or cyanuric acid mixture on the sperm quality of male mice in an acute experiment.42 male SPF KM mice were divided into 4 groups randomly.Melamine and cyanuric acid were mixed at the ratio of 1:1.Then the mice from three group were administered instagastrically with low dosage(30mg/kg.bw.d), moderate dosage (140mg/kg.bw.d) and high dosage(700mg/kg.bw.d), respectively for 3 days Simultaneously,the control mice were administered with equivalent corn oil. At 24h after the last administration,the blood was collected and then all the mice were sacrificed.Prepare the sperm suspension,count the quantity of the sperm,the sperm motility rate and the sperm abnormality rate.Detect the sperm DNA damage by single cell gel electrophoresis.The results showed that high doses of MA-CA can cause a significant decrease in the number of sperm in epididymis.MA and MA-CA in each dose group significantly reduced the living sperm rate, significantly increased the rate of sperm deformity, especially the head deformity rate, and showed a dose effect.High dose of MA and MA-CA caused significant damage to sperm DNA.These results indicate that MA and MA-CA in acute test have obvious damage to sperm quality in male mice, and MA-CA is more serious damage.
