长链非编码RNA-HULC对结直肠癌细胞生长和转移的作用
Effects of LncRNA-HULC on proliferation and metastasis of CRCcells
目的：研究长链非编码RNA（long noncoding RNA，LncRNA）HULC对结直肠癌（colorectal cancer，CRC）细胞增殖、侵袭、凋亡的作用。方法：通过si-RNA干扰技术构建HULC低表达CRC细胞系，下调HULC表达水平后，用BrdU染色实验、Transwell侵袭实验以及Tunel染色检测HULC对CRC细胞的影响。结果：Brdu染色提示HULC可促进CRC细胞增殖，Tunel染色提示HULC可显著抑制CRC细胞的凋亡能力，Transwell侵袭实验发现 HULC能显著促进CRC细胞的侵袭能力。结论：LncRNA-HULC表达可以促进结直肠癌细胞的生长、侵袭，并抑制凋亡。
Objective: To evaluate effects of long noncoding RNA (lncRNA) HULC on proliferation and invasion of colorectal cancer (CRC) cells in vitro. Methods: The HULC siRNA was constructed and respectively transfected into CRC cell with low expression of HULC. After down-regulation of HULC expression, cellular proliferation inhibitory activity was determined by BrdU assay. Transwell invasion assay was used. Additionally, the apoptosis activity was tested by tunel assay. Results: BrdU assay indicates knockdown of HULC represses CRC cell proliferation. Transwell invasion assays reveal knockdown of HULC represses CRC cell invasion. Tunel assay shows knockdown of HULC facilitates CRC cell apoptosis. Conclusion: HULC promotes CRC cell proliferation, invasion, and induces cell apoptosis in vitro.
