不同基质上绿僵菌孢子萌发及附着孢观察

孢子萌发及附着孢形成是昆虫病原真菌侵染寄主过程中的关键步骤。本文对金龟子绿僵菌CQMa102菌株孢子萌发及附着孢形成率、芽管及附着孢形态在不同基质上孢子萌发及附着孢形成率、芽管及附着孢形态进行了研究。结果表明在玻璃基质上营养液的丰富程度、培养时间对孢子萌发和附着孢形成均具有显著影响。高浓度营养液有利于孢子萌发,但不利于附着孢形成。附着孢通常在培养20-24h时出现,24-48h内大量生成。与用玻璃作基质相比,蝗翅有利于诱导附着孢形成,且蝗翅上绿僵菌附着孢个体较大,芽管短、分枝少,隔膜少,附着孢形成均一性好。
onidium germination and appressorium formation are key steps in the process of infecting insect hosts by entomopathogenic fungi. Growth media with four different nutrition levels combined with various cultivation intervals were utilized to study the effect on conidial germination and appressorial development, as well as properties of germ tube and appressorium of Metarhizium anisopliae, CQMa102, when applied on glass surface and locust wing. Results showed that nutrition level of the growth media and cultivation interval significantly affected conidial germination and appressorial formation. Nutrient-rich media enhanced conidial germination but negatively impacted appresorial formation. Appressoria were formed mostly during the period of 24 - 48h post cultivation. Compared with glass surface, there were more appressoria produced on locust wing. Moreover, larger sized appressoria with germ tubes having shorter length, fewer branches, and fewer septa were found on locust wing than those on the glass surface.
