摘要

本文应用免疫组织化学法研究了大鼠全脑缺血后,海马CA1、CA3区中GLT-1及GFAP 的表达。15只雄性Wistar大鼠,随机分为3组:control组;假手术组;损伤性缺血组。假手术组、损伤性缺血组大鼠分别在假手术后或缺血再灌后常温饲养7天。损伤性缺血组,海马CA1区锥体神经元几乎全部死亡;死亡的锥体细胞周围GLT-1的表达减少,甚至表现为GLT-1大片缺失;同时可见该区星型胶质细胞的胞体肥大,突起增粗、但明显断裂、残缺。损伤性缺血组海马CA3区的神经元未见到明显的损伤;GLT-1的表现明显上调,尤其以锥体细胞周围上调的最为明显,在锥体细胞周围形成非常明显的网格状结构;该区星型胶质细胞的胞体并不明显增大,但突起明显环绕神经元胞体,形成非常明显的网格状。提示,海马CA3区锥体神经元周围大量表达的GLT-1有利于保护锥体神经元免于缺血性损伤。

Abstract

The expression of GLT-1 and GFAP in the hippocampal CA1 and CA3 subfield after global brain ischemia was studied by immunohistochemistry in the experiment. Fifteen adult male Wistar rats were randomly divided into 3 groups: control group; sham group and global brain ischemia group. Rats in sham group and global brain ischemia group were raised for 7 days after the sham operation or reperfusion. In the hippocampal CA1 subfield after global brain ischemia insult, almost all pyramidal neurons died. The GLT-1 expression was markedly decreased in the area where almost complete pyramidal neurons died, and the neighboring area of the pyramidal layer even appeared as a sheet absence of GLT-1 immunoreactivity. At the same time, astrocytes hypertrophied in soma with thickened processes that began to collapse and fragment. In the hippocampal CA3 subfield after global brain ischemia insult, no obvious pyramidal neuronal injury was observed. Many GLT-1 immunoreactive particles were observed in the area between the pyramidal neurons, which tightly surrounded pyramidal neurons and made the pyramidal layer look like ‘shaped grade’. The cell body of astrocytes was not hypertrophy. Some GFAP immunoreactive particles were observed in the area between the pyramidal neurons, which tightly surrounded the pyramidal neurons and made the pyramidal layer look like a ‘shaped grade’. All the above suggested that the GLT-1 expression surrounded pyramidal neurons in the hippocampal CA3 subfield protected the neurons survival the global brain ischemic insult.
