小鼠乳腺中Heregulin-α的信号转导通路
Study on signal transduction pathway of Heregulin-α in mammary gland of mous
为研究小鼠乳腺发育、泌乳及退化过程中Heregulin-α的信号转导通路,分别取妊娠9 d、泌乳12d 以及退化6d 小鼠乳腺组织进行体外培养,对照组培养基为无血清培养基,处理组培养基中则只添加Heregulin-α。培养24h后分别提取乳腺上皮细胞总蛋白,用免疫印迹法检测对照组和处理组乳腺上皮细胞中信号分子STAT3、STAT5、p44/42 MAPK和p38 MAPK的磷酸化情况,并通过ELISA方法检测对照组和处理组乳腺上皮细胞中信号分子PKC的磷酸化情况。结果表明, Heregulin-α能够促进妊娠期小鼠乳腺上皮细胞p44/42 MAPK、p38 MAPK和PKC的磷酸化; Heregulin-α能够促进泌乳期小鼠乳腺上皮细胞STAT5和p38 MAPK的磷酸化,而同时又抑制PKC的磷酸化; Heregulin-α能够增强退化期小鼠乳腺上皮细胞STAT3的磷酸化,并抑制PKC的磷酸化。
o determine the signal transduction pathway of Heregulin-α in mammary gland, the mammary gland of mouse at 8 days in pregnancy, 12 days in lactation and 6 days in involution were cultured in vitro. Each individual mammary gland was cultured in the media Waymouth′s MB752/1 with or without Heregulin-α. Following 24 hours of culture, each mammary gland was digested to obtain epithelial cells, which were lysed and the lysate was used for western blot analysis. Western blot results showed that Heregulin-α enhanced the phosphorylation of p44/42 MAPK, p38 MAPK and PKC in pregnancy; and, Heregulin-α promoted the phosphorylation of STAT5 and p38 MAPK, while it inhibited PKC phosphorylation in lactation; in involution, Heregulin-α induced STAT3 phosphorylation and inhibited PKC phosphorylation.
