目的:制备硫酸化茶藨子木层孔菌多糖,研究其体外的抗肿瘤活性,并与茶藨子木层孔菌多糖比较。方法 采用氯磺酸-甲酰胺法进行硫酸化,采用MTT法检测对人卵巢癌细胞SKOV-3的影响。结果:制备了3种茶藨子木层孔菌多糖硫酸酯,硫取代度分别为1.48、1.83和2.23,其红外光谱中均具有S=O和C-O-S的伸缩振动峰。茶藨子木层孔菌多糖对SKOV-3细胞无明显的影响,而其硫酸酯可以抑制SKOV-3的生长。结论:成功制备了没有降解的3种硫酸酯,茶藨子木层孔菌多糖经硫酸化后对于肿瘤细胞的抑制作用增强。

Objective To prepare sulfated polysaccharide from Phellinus ribis (PRP-S) and study their antitumor activities compared with Phellinus ribis polysaccharide (PRP) in vitro. Method PRP-S was prepared by chlorosulfonic acid- formamide method. The effects on human ovary cancer SKOV-3 cells were observed in vitro by MTT method. Result: The degrees of sulfate of three sulfated derivatives (PRP-S) were 1.48, 1.83 and 2.23, respectively. Two characteristic absorption bands (S=O and C-O-S) appeared in FT-IR spectra. PRP had no obvious influence on SKOV-3 cells, however PRP-S exhibited much stronger inhibition effects against SKOV-3 cell growth. Conclusion: PRP-S was obtained successfully without degradation of the polysaccharide. PRP-S showed significant inhibition effects on SKOV-3 cells in comparison with the native non-sulfated polysaccharide.
