Genetic Stability of cpDNA in Long-term Sub-cultured Citrus Callus

对23份经过7—20年保存的柑橘愈伤组织的质体基因组(叶绿体cpDNA)的遗传稳定性进行了RFLP和叶绿体SSR(cpSSR)分析。RFLP分析结果表明愈伤组织在保存过程中cpDNA的含量无明显变化;但cpSSR分析发现引物对SPCC4在愈伤组织中检测到比叶片多了一条谱带,Southern杂交和序列测定均表明,该谱带为特异扩增产物,说明愈伤组中质体基因组普遍存在短核苷酸序列插入现象。

Restriction fragment length polymorphism (RFLP) and chloroplast simple sequence repeats (cpSSR) were employed to assay the chloroplast genome (cpDNA) genetic stabilities in long-term sub-cultured citrus callus. Without any significant difference was observed by RFLPs, however, a novel PCR fragment was obtained by SPCC4 primer pairs. Further Southern blotting and sequencing proved that this fragment was specifically amplified, which indicated that short insertion occurred in the cpDNA of Citrus calluses.
