目的:阐明飞蝗羧酸酯酶基因在溴氰菊酯作用下mRNA表达特性。方法:利用不同剂量溴氰菊酯处理飞蝗,提取总RNA,体外反转录合成cDNA模板,采用Real-time PCR技术分析飞蝗羧酸酯酶基因在溴氰菊酯不同浓度和不同时间处理后的表达模式。结果:飞蝗经不同浓度溴氰菊酯处理12 h后,部分羧酸酯酶基因mRNA表达量受到抑制;除LmCesA3和LmCesE1外,其余羧酸酯酶基因经溴氰菊酯处理不同时间后表现为先诱导而后抑制。结论:溴氰菊酯对飞蝗不同羧酸酯酶基因mRNA表达的影响有所差异,研究结果将为揭示飞蝗抗药性机理提供理论依据。

Objective: To analyze the mRNA expression patterns of carboxylesterase genes of L. migratoria to deltamethrin exposures. Methods: locusts exposed to different doses of deltamethrin and treated with deltamethrin at different exposure times were collected as templates, respectively. cDNAs were synthesized from total RNA of different templates using reverse transcriptase. Further, mRNA expression response of carboxylesterase genes of L. migratoria to deltamethrin exposures were confirmed by Real-time PCR. Results, exposures of deltamethrin at different doses for 12 h suppressed the mRNA expressions of most carboxylesterase genes. Except for LmCesA3 and LmCesE1, the transcript levels of most carboxylesterase genes were induced at early treated time and were suppressed at later treated time when the locusts were treated with deltamethrin at different exposure times. Conclusion: deltamethrin influenced the expression of carboxylesterase genes, ranging from induction to repression. The results will be helpful in providing strategies for insecticide resistance management of L. migratoria.
