酿酒酵母菌中脂滴结构维系因子筛选
Screening of structural maintenance factors for lipid droplets in Saccharomyces cerevisiae
脂滴存在于所有真核生物和部分原核生物中,是细胞内调节脂质代谢与能量代谢的重要细胞器之一。酿酒酵母的细胞中通常存在多个脂滴,且脂滴大小相对均一、结构和数量相对稳定。即便处于在极短的时间内需快速消耗或积累大量中性脂的极端条件下,脂滴也极少出现彼此融合的现象;然而酵母细胞维系成熟脂滴的结构稳态机制却知之甚少。本研究通过油酸培养对酿酒酵母的脂滴表面蛋白进行筛选,寻找维持脂滴结构稳定性的作用因子,结果发现Pln1p对于脂滴维持结构的相对稳定至关重要。
Lipid droplets are present in all eukaryotes and some prokaryotes, and are one of the important organelles regulating lipid and energy metabolism in cells.In Saccharomyces cerevisiae, multiple lipid droplets are usually present in the cell, and the droplets are relatively homogeneous in size and stable in structure and number. Even under extreme conditions where large amounts of neutral lipids are rapidly consumed or accumulated in a very short period of time, lipid droplets rarely fuse with each other; however, the structural homeostatic mechanisms by which yeast cells maintain mature lipid droplets are poorly understood. In this study, we screened the surface proteins of lipid droplets of Saccharomyces cerevisiae by oleic acid incubation to search for the role of factors that maintain the structural stability of lipid droplets, and found that Pln1p is essential for the maintenance of relative structural stability of lipid droplets.
