水稻苗期耐冷相关QTL qCTS3-1的鉴定和分子定位
Identification and Mapping of a Major QTL qCTS3-1 Controlling Cold Tolerance at Seeding Stage in Rice
本研究以广陆矮4号为受体,日本晴为供体的染色体片段代换系群体中的1个低温敏感株系C32022为材料,以 低温处理后植株存活率为指标,采用代换作图法在水稻第3染色体上定位了一个控制水稻苗期耐冷性的QTL,命名为qCTS3-1。利用C32022和广陆矮4号衍生的F2分离群体进行遗传分析,结果表明,该群体中叶片表现正常绿色和叶片表现出黄色、萎蔫单株比例符合1 : 3的分离比,表明耐冷性受单个主效QTL控制,且低温敏感对低温耐受为显性来自日本晴的等位基因增加对低温的敏感性。利用该群体中86个隐性单株将qCTS3-1初步定位在区WZD-S3-7~WZD-S3-11之间,物理距离约为550kb。研究结果为qCTS3-1的精细定位和克隆奠定了基础。
In this study, we characterized a cold tolerance sensitive line C32022 from a set of chromosome segment substitution lines derived from a cross between Guangluai 4 as the recurrent parent and Nipponbare as the donor. Survival rate after cold treatment was selected as the screening index for cold tolerance at seedling stage. A major QTL, qCTS3-1, was mapped on chromosome 3 using substitution mapping strategy. In an F2 segregating population consisting of 401 plants derived from C32022 and Guangluai 4, the frequency distribution of qCTS3-1 was shown to be an expected Mendelian ratio for single locus segregation. Segregation analysis also indicated that the cold sensitive allele came from Nipponbare. The qCTS3-1 locus was further narrowed down to approximately 550-kb DNA fragment defined by newly developed STS markers WZD-S3-7 and WZD-S3-11 using 86 recessive individuals. These results provide useful information for fine mapping and cloning of qCTS3-1.
