目的:探讨小鼠成神经母细胞瘤细胞(N2a)不同细胞周期中线粒体分裂蛋白(Fis1)、线粒体融合蛋白(Mfn2)及微管相关蛋白1轻链3(LC3)表达水平的变化。方法:通过胸腺嘧啶核苷(thymidine)和诺考达唑(nocodazole)干预,使N2a同步化在不同细胞周期(G1/S期、S期、G2/M期);流式细胞仪鉴定细胞周期;Western blot检测Fis1、Mfn2、LC3在不同细胞周期中的表达水平。结果:(1)流式细胞仪检测各组DNA含量百分比显示G0/G1、S、G2/M期均同步化,符合本实验要求。(2)Western Blot示:同步化干预后,Mfn2和LC3在G0/G1、G2/M期的表达量显著低于S期(p<0.01 或p<0.05);Fis1在G0/G1、S、G2/M各期表达不同,S、G2/M期表达量高于G0/G1期,但不具有统计学意义。结论:Fis1、Mfn2、LC3的表达随着细胞周期的变化而变化,Mfn2、LC3在S的表达明显高于G0/G1、G2/M期。
Objective: To evaluate the change of mitofusin2 (Mfn2), mitofision1 (Fis1), microtubule-associated protein1 light chain3 (LC3) expression in the different phases of cell cycle of mouse neuroblastoma cells (N2a) in vitro. Methods: double- thymidine block was introduced to synchronize cells in the different phases of cell cycle (G0/G1- phase, S- phase, G2/M-phase) and flow cytometry was used for identification. The expression of Mfn2, Fis1, LC3 were detected by Western-blot. Results: (1) Flow cytometry revealed that cells were synchronized in G0/G1- phase, S- phase and G2/M-phase, respectively. (2) The expression level of Mfn2 and LC3 in S-phase were significantly higher than those in G0/G1 and G2/M-phase. The expression level of Fis1 was different in these three phases, but the difference is not significant. Conclusion: The expression of Mfn2, Fis1, LC3 change with the processing of cell cycle, and the expression levels of Mfn2 and LC3 were higher in S-phase than it in G0/G1 and G2/M-phase.
