加减青娥方体外对RAW264.7细胞向破骨细胞分化的影响
Effect of Modiffied Qing E Formula in menopausal osteoporosis differentiated from RAW264.7 in vitro
目的:探讨加减青娥方对RAW264.7向破骨细胞分化的影响。方法:以体外细胞培养的方式,在小鼠源性破骨前体细胞RAW264.7细胞中加入不同浓度的加减青娥方提取物进行干预,MTT法检测不同浓度加减青娥方对RAW264.7细胞增殖的影响,TRAP染色法检测不同浓度的加减青娥方对RAW264.7细胞向破骨细胞分化的影响。结果:不同浓度加减青娥方均可以减少RANKL诱导的破骨细胞的数量,其作用与17β-雌二醇效果相似。结论:青娥方可抑制破骨细胞的分化、增殖,从而实现增加骨密度、防治骨质疏松的作用。
Objectives: The aim of this study was to explore the effect of Modified Qing E Formula on the osteoclast differentiation from osteoclast precursors RAW264.7 cells. Materials and Methods: The extractions of Modified Qing E Formula with different doses were added into the osteoclast precursors RAW264.7 cultured in vitro. Then the effects of the Modified Qing E Formula on the generation of the RAW264.7 cell,the number and activity of osteoclasts differentiated from osteoclast precursors RAW264.7 induced by receptor activator of nuclear factor κ B factor ligand (RANKL) were detected with tartrate-resistant phosphatase (TRAP) staining. Results: Modified Qing E Formula inhibited the RANKL-induced RAW264.7 cells to differentiate into osteoclasts,same as the action of 17β-estradiol. Conclusions: Modified Qing E Formula could increase bone density to prevent and treat PMOP via suppressing the differentiation and proliferation of osteoclasts.??
