目的 研究慢病毒介导COX-2基因沉默对Tca8113细胞株生长及侵袭转移能力的影响。方法 通过前期实验室建立的稳定沉默COX-2的Tca8113细胞系本实验设COX-2基因沉默组(pLKO.1shCOX-2)、空载体载体阴性对照组( Tca8113 scr)、正常组(Tca8113)。以上三组细胞分别采用western-blot检测COX-2蛋白的表达情况,通过细胞克隆形成实验检测慢病毒介导COX-2基因沉默后对Tca8113细胞成瘤情况的影响,通过Transwell小室,检测COX-2基因沉默后对Tca8113细胞侵袭转移能力的变化。结果 western-blot结果显示,pLKO.1shCOX-2基因沉默后较Tca8113 scr组,Tca8113组蛋白表达明显降低;COX-2基因沉默后Tca8113细胞成瘤性降低,侵袭转移能力减弱。结论 经慢病毒介导COX-2基因沉默能够抑制Tca8113细胞株的成瘤性及侵袭转移能力。

Objective To explore the effect of lentivirus-mediated COX-2 on the proliferation and invasion of Tca8113. Methods The frist step was to establish the stable Tca8113 cell lines with silent COX-2. Then to set up lentivirus-mediated COX-2 gene silencing experimental group (pLKO.1shCOX-2), non-carrier negative control group (Tca8113 scr) and normal cells blank control group (Tca8113). Results The western-blot was used to measure the expression of COX-2 protein and the cloning formation assay was used to examine the oncogenicity of Tca8113 impacted by silent lentivirus-mediated COX-2 gene. The changes of Tca8113 in invasion and metastasis were detected by Transwell chamber.Conclusion Down-regulation of COX-2 expression can inhibit the tumorigenicity Tca8113,and it can weaken invasion and etastasis of Tca8113 cells.
