Sp1-like proteins and Krüppel-like factors belong to the zinc-finger protein family with multiple members and similar structures, which play the important role in the gene transcriptional regulation of eukaryote cells. For homo sapiens, there are at least 21 kinds of Sp1-like/KLF proteins. KLF4 is a highlight of KLF family members. It can bind with the KLF4 binding element on promoters of target genes to control the transcription of target genes, which is an important transcriptional factor. It has been demonstrated that KLF4 plays the important roles in various physiological and pathological processes, including cell proliferation and differentiation, tumorogenesis and vascular reconstruction. This paper reviews the structure of Sp1-like/KLF protein family, the structural properties of KLF4 and the biological functions of KLF4.
