棉花GAUT家族的鉴定与表达分析

Identification and expression analysis of GAUT family genes in cotton

棉花是我国最重要纤维经济作物,果胶在棉花的生长发育过程中扮演着至关重要的作用。半乳糖醛酸转移酶(Galacturonosyltransferase,GAUT)是一种参与催化糖基化反应的酶类,也是果胶代谢途径上的关键酶,其功能具有多样性。本研究基于棉花现代栽培种陆地棉NDM8和海岛棉Pima90的基因组数据,通过生物信息学手段发现陆地棉和海岛棉分别具有40和41个GAUT家族成员;系统进化分析显示棉花GAUT家族成员与拟南芥GAUT家族成员具有同源性。共线性分析显示陆地棉和海岛棉中的大多数GAUT家族成员之间大多存在共线性关系,其中GhGAUT22与GbGAUT23受到正选择,其他成员受到纯化选择。对陆地棉GAUT家族成员的表达分析发现:GAUT家族成员具有时空表达特性;且对不同的胁迫响应存在显著的差异,其中对干旱胁迫响应最为明显,位于多个显著上调表达的成员启动子区域都具有一个GT-1元件,但不存在于多个下调趋势较大的成员启动子中。qRT-PCR结果显示陆地棉和海岛棉各3个基因在不同组织的表达水平与转录组数据相近。本研究从全基因组水平鉴定了棉花现代栽培种的GAUT家族成员并进行生物信息学和表达模式分析,为研究GAUT基因的分子生物学功能提供了理论基础。

otton is the most important fiber cash crop in China. Pectin plays an important role in the growth and development of cotton. Galacturonic acid transferase (Galacturonosyltransferase, GAUT) is a kind of enzymes involved in catalytic glycosylation, also is the key enzyme of pectin on the metabolic pathway, its function has diversity. In this study, based on genomic data of modern cultivated cotton species upland cotton NDM8 and island cotton Pima90, it was found that upland cotton and island cotton had 40 and 41 GAUT family members, respectively. Phylogenetic analysis showed that GAUT family members of cotton and Arabidopsis had homology. Collinearity analysis showed that there were collinearity relationships between most GAUT family members in upland cotton and sea island cotton, in which GhGAUT22 and GbGAUT23 were selected positively, and other members were selected purified. The expression analysis of GAUT family members in upland cotton showed that GAUT family members had temporal and spatial expression characteristics. There were significant differences in response to different stress, among which the response to drought stress was the most obvious. GT-1 element was located in several member promoter regions with significantly up-regulated expression, but it did not exist in multiple member promoters with a large down-regulated trend. qRT-PCR results showed that the expression levels of three genes in upland cotton and sea island cotton were similar to the transcriptome data. In this study, GAUT family members of modern cultivated cotton species were identified from the whole genome level and bioinformatics and expression patterns were analyzed, which provided the basic theoretical basis for studying the molecular biological function of GAUT gene.
