针刺调整下丘脑-垂体-卵巢轴异常功能的物质基础研究

Study of Effective Substances in Elector-acupuncture Normalizing the Dysfunction of Hypthalamus-Pituitary-Ovary axis

本文采用下丘脑视前区(MPO)灌流液异体转移的方法,观察针刺疗法的病理生理机制。将有正常性周期的雌性大鼠随机分成四组,分别收集动情前期灌流液组(PEP),动情期灌流液组(EP);动情后期灌流液组(MEP)和动情间期灌流液组(DEP)。将去卵巢的雌性大鼠分为收集电针后下丘脑MPOA推挽灌流液组(OVXAP);去卵巢大鼠MPOA灌流液组(OVXP);注射针刺灌流液组(IOVXAP);注射去卵巢灌流液组(IOVXP);注射动情前期灌流液液组(IPEP)和注射人工脑脊液对照组(IaCSF)。IOVXAP大鼠的血E2显著升高,阴道涂片成熟脱落细胞重新出现,而IOVXP和IaCSF未出现类似的变化;正常大鼠性周期四个阶段的灌流液中的神经地质的变化与GnRH的变化相一致。IPEP大鼠的阴道脱落细胞重新出现,血雌激素水平较升高。而OVXP组未见相似变化。结果提示针刺对病理变化的调节亦是通过影响下丘脑这些生物活性物质实现的。灌流液转移作为一种研究方法,为以后开展离体研究针刺效应的机理提供一种新的实验方法。

In experiemt 1, the push-pull perfusion in hypothalamic preoptic area (MPO) of the ovariectomiezed rats treated with electroacupuncture (name as EA perfusate) was transferred into the MPO of another ovarictomized rat without EA treatment. The blood E2 level in the recipient increased significantly and a few of mature vaginal epithelia reappeared. Those changes were not observed in recipient with perfusate from ovariectomized rat or artifical cerebrospinal fluid. In experiment 2, the push-pull perfusate in hypothalamic preoptic area (MPO) of the rat in different estrus cycle was collected and determined the concentration of neuroactive substances in those perfusate with HPLC and RIA. Then perfusate from rat in pre-estrous cycle (PEP) was microinjected into the MPO of the ovareicetomized rats. The blood E2 level in the ovariectomized rat with PEP microinjection increased significantly compared with the ovariectomized rat with OVXP. A few of mature vaginal epithelia reappeared in ovariecotmized rat with PEP microinjection. Present data suggested that neuroactive substances in MPO were the effective substances of physiological regulation function and pathological changes. They are also the effective substances in electoracupuncture treatment.
