家蚕S期激酶相关蛋白的克隆表达和定位研究

家蚕S期激酶相关蛋白是一类泛素化蛋白,主要参与细胞信号传导、细胞分裂与分化等多种重要的生物过程。从本实验室构建的cDNA文库中克隆到一条新的基因序列,经blastn序列比对后,推测是家蚕S期激酶相关蛋白(S-phase kinase associated protein,Skp ),命名为BmSkp。构建重组表达质粒pET-28a-BmSkp,在大肠杆菌BL21(DE3)中诱导表达。亲和层析纯化目的蛋白His-BmSkp。以纯化的融合蛋白为抗原免疫雄性新西兰兔制备多克隆抗体,间接ELISA方法检测抗体效价可达1:12800,Western blot检测表明制备的多克隆抗体有较高的特异性。用荧光定量PCR和Western blot实验分析BmSkp基因在家蚕不同时期和不同组织中的转录差异和表达情况。亚细胞定位研究还表明BmSkp蛋白主要存在于细胞质中,为BmSkp白蛋功能的研究进一步奠定了基础。
Skp (S-phase kinase associated protein) was a kind of F-box protein which participated in the ubiquitination. A cDNA sequence containing three Skp1 conservative domains was cloned in a Bombyx mori cDNA library, and designated as BmSkp since it encoded an S phase kinase-associated protein of silkworm. This gene was inserted into a vector pET-28a with both EcoR I and Xho I restriction sites, and then expressed in E. coli BL21 (DE3). The recombinant protein was determined to be 26 kD by the SDS-PAGE analysis. The target protein was purified by affinity chromatography and used to immune a male New Zealand rabbit for the production of anti-serum. The polyclonal antibody was generated and the titer was 1:128,000 measuring by ELISA and was determined for specificity by Western blot after purified by affinity chromatography using immobilized protein A. The polyclonal antibody was generated and the titer was 1:21800 measuring by ELISA and was determined for specificity by Western blot after purified by affinity chromatography using immobilized protein A. The total proteins and RNA from four developmental stages and eight different tissues of silkworm were extracted for real-time PCR and Western blot, in order to analysis the transcription and expression level of BmSkp. Subcellular localization of BmSkp was shown to be mainly present in cytoplasm of the BmN cells. These researches may lay a foundation for the study of BmSkp gene function.
