目的:探讨Garcinol对DMBA诱导地鼠颊癌模型早期炎症的影响。方法:27只金黄地鼠雄性,6-8周龄。3只不涂药作为阴性对照,其余24只于左侧颊囊涂0.5%DMBA,每周3次, 共涂3周。3周后随机分为4组,阳性对照组不涂药,其余3组地鼠于左侧颊囊分别涂0.5mM、5mM、50mMGarcinol,一周涂3次。第4周末实验结束处死动物,取左侧颊囊进行组织病理学观察和BrdU免疫组化染色。同时取小部分组织冰冻,以进一步分析PGE2和LTB4。结果:与阳性对照组相比,各浓度Garcinol组抑制了单纯增生、异常增生灶的数目(P< 0.05);每平方毫米炎症细胞的数量显著减少(P<0.01);中、高浓度 Garcinol组BrdU增殖指数显著降低(P<0.05);与阳性对照组相比,低、中、高浓度Garcinol组的PGE2水平未见明显差异(P>0.05);不同浓度Garcinol组LTB4水平显著降低(P<0.01)。结论:本研究显示Garcinol对DMBA地鼠模型早期炎症的有一定影响,可以减少炎症细胞数目,抑制细胞增殖。

Objective:To evaluate the anti-inflammatory effect of Garcinol on DMBA-induced hamster cheek pouch inflammation model. Materials and Methods: The hamsters were divided into five groups. While the negative control group was not treated, the other Syrian hamsters were treated with 0.5% DMBA solution topically to the left cheek pouch three times per week for three consecutive weeks. After the last treatment of DMBA, the hamsters were divided into four groups at random. The positive group was not treated, the other three groups received 0.5mM、5mM、50mM Garcinol respectively three times within the following week. Hamsters were sacrificed at the forth week to obtain tissue samples of the left cheek pouch. They were injected with 5-bromodeoxyuridine (BrdU) 50mg/kg before they were sacrificed . A piece of tissue from each hamster was snap-frozen in liquid nitrogen for PGE2 and LTB4 ELISA analysis. The remaining samples were examined by histopathology and BrdU immunohistochemisty. Results: 1) The number of oral lesions (hyperplasia and dysplasia) decreased after 0.5mM, 5mM, 50mM Garcinol treatment(P<0.05); 2) 0.5mM, 5mM and 50mM Garcinol treatment significantly decreased the number of inflammatory cells/mm2 from 320.51±155.88 to 197.72±83.77, 184.39±64.16,100.41±52.53, respectively(P<0.01); 3) Moderate and high doses of Garcinol decreased the indices of BrdU from 4.26±2.21% to 3.30±1.37% (P<0.05) , 3.01±1.78% (P<0.01) ; 4)With ELISA, we observed that the content of PGE2 was of no significant difference between groups. The LTB4 level of high dose of Garcinol group was significantly decreased compared with positive control(P<0.01). Conclusions: These results indicated that Garcinol have anti-inflammatory, and cell proliferation suppressing effect in DMBA-induced inflammation of hamster cheek pouch.
