miR-223通过抑制IGF-1R信号通路诱导人血管平滑肌细胞自噬
MicroRNA-223 induces autophagy of human vascular smooth muscle cells by inhibiting IGF-1R signaling pathway
目的探究miR-223对人动脉平滑肌细胞(HASMCs)自噬功能调控的分子机制。方法收集正常捐献者的下肢动脉标本并提取HASMCs,通过RT-qPCR、免疫印迹反应实验观察比较实验组(转染miR-223 mimic)和阴性对照组IGF-1R-PI3K/Akt信号通路表达差异以及自噬相关蛋白表达差异。结果转染miR-223 mimic的HASMCs与阴性对照组相比,IGF-1R-PI3K/Akt信号通路明显下调,Western blotting结果表明自噬功能明显增强。结论 miR-223在HASMCs中通过与靶基因IGF-1R mRNA的3\'-UTR直接结合,降解mRNA并抑制下游PI3K/Akt信号传导通路的激活,导致HASMCs细胞自噬水平的明显升高。?????
Purpose To explore the molecular mechanism of miR-223 on the autophagy function of human artery smooth muscle cells (HASMCs) Methods Donors\' normal lower limb arteries were collected and HASMCs were extracted. By RT-qPCR and western blot, differences of IGF-1R-PI3K/Akt signaling pathway expression and autophagy related protein expression between experimental group (miR-223 mimic transfected) and negative control group were observed. Results HASMCs transfected with miR-223 mimic showed a significant down-regulation in the IGF-1R-PI3K/Akt signaling pathway compared with the negative control group, with significant enhancement of autophagy flux. Conclusion miR-223 binds IGF-1R mRNA 3 \'UTR in HASMCs, which leads to degradation of the mRNA and suppression of the activation of IGF-1R-PI3K/Akt signaling pathway, and up-regulates HASMCs\' autophagy flux.
