目的 基于敲除敲入的模式,研究金银花中清除氧自由基的活性组分。方法 采用大孔树脂和聚酰胺层析柱富集各个组分,以各组分对活性氧自由基(DPPH•)的清除活性为评价指标,确定药效组分。结果 DPPH•清除能力:有机酸IC50= 0.109mg/ml,总黄酮IC50=0.129mg/ml,两者分别具有对DPPH自由基清除的能力;环烯醚萜苷IC50=1.097mg/ml,总皂苷IC50=1.158mg/ml,两者对DPPH自由基的清除能力很弱或不具有清除活性。 结论 金银花中具有抗氧化作用的药效组分为有机酸类及黄酮类成分,该模式能用于中药药效物质的辨识。

IM To study Antioxidant constituents of Lonicera japonicaThunb. based on the model of “knock-out & knock-in”. METHODS Active components were concentrated by macroporous resin column and polyamide column.The components were detected by DPPH•free radical-scavenging method. RESULTS DPPH• Scavenging ability: organic acids part IC50= 0.109mg/ml, total flavonoids IC50=0.129 mg/ml, both of them have the ability to scavenge DPPH•free radical; Iridoid glucosides IC50=1.097 mg/ml, total saponins IC50=1.158mg/ml, the ability to scavenge DPPH•free radical of them were weak. CONCLUSION Antioxidant constituents from flower of Lonicera japonicaThunb. were organic acids and flavonoids. This model could be used to recognize the pharmacodyamic constituents of Traditional Chinese Medicine.
