复合微球的聚乳酸静电纺丝纤维作为药物载体的研究
Investigation of Electrospinning PLLA Fiber Composited with Chitosan Microspheres for Drug Delivery
聚乳酸静电纺丝纤维作为药物载体已经被广泛的研究,但是对于一些水溶性的药物分子的负载率低。本研究以水溶性的牛血清白蛋白为模板药物,通过喷雾干燥的方法制备了负载蛋白的壳聚糖微球,该微球的粒径在2-4μm之间。将微球通过超声分散在聚乳酸的溶液中,通过静电纺丝技术,制备出复合载药微球的聚乳酸超细纤维,然后研究该复合纤维的形貌、微球分布,并且通过加入聚乙烯吡咯烷酮(PVP)来调节药物释放性能。结果表明,载药微球能够在聚乳酸超细纤维中分布均匀,体外释放表明其具有良好的药物释放性能,PVP的加入可以明显加快其释放性能。这种新型的复合微球的聚乳酸静电纺丝纤维能够较好的负载和释放水溶性的药物。
Electrospinning PLLA fiber as a drug carrier has been extensively studied, however, it has a low load efficiency for some water-soluble drug molecules. In this paper, microspheres loaded with water-soluble BSA were prepared by spray drying, and then microspheres were dispersed in PLLA solution. The PLLA microfiber composited drug-loaded microsphere was prepared through the electrospinning technique. The morphology of the composite fiber was studied, and the drug release performance was regulated by adding polyvinylpyrrolidone (PVP). The results showed that the distribution of drug-loaded microspheres in ultra-fine fibers of polylactic acid was uniformity. The release in vitro showed that it has a good drug release properties, and the addition of PVP can significantly speed up its release.
