马尾松胚性细胞悬浮增殖培养体系的建立

Establishment of embryogenic suspension cultures in somatic embryogenesis of Pinus massoniana Lamb.

本研究旨在利用马尾松未成熟胚诱导的胚性愈伤组织,建立马尾松胚性愈伤组织悬浮增殖培养体系。论文分别对悬浮细胞生长参数沉淀细胞体积(SCV)、细胞活力指标进行了测定,研究初始接种量,继代细胞密度对悬浮培养增殖的影响以及悬浮细胞增殖培养的生长和细胞活力的动态变化。研究结果表明:不同初始接种量、继代细胞密度显著地影响马尾松胚性细胞的悬浮增殖培养;在20天的悬浮增殖培养过程中,胚性细胞的生长周期曲线呈"S"型,细胞活力呈现先急剧上升,后缓慢下降的趋势。本研究首次建立了最适的马尾松胚性愈伤悬浮增殖体系,即在30ml的液体培养基中,初始接种1.0g胚性愈伤组织,悬浮培养三周后,平均增殖系数可达12.097,其后继代培养以1/6体积比作为转接继代培养细胞密度参考值,适宜的继代间隔期为8-12d。本研究为了解马尾松胚性细胞悬浮增殖培养的生长规律,优化调控马尾松体细胞胚胎发生快繁增殖体系提供了技术支撑,同时为实施马尾松基因工程遗传转化以及采用细胞融合进行马尾松新品种选育等奠定了研究基础。

Suspension cultures were firstly established with embryogenic tissues initiated from immature zygotic embryos in Pinus massoniana Lamb. (masson pine ). The effects of initial tissues weight, cell density in subculture , the dynamic changes of cell growth and cell viability were investigated through measuring the sediment cell volume (SCV) and cell viability by spectrophotometer during establishment of suspension cultures. The results showed that: the growth of embryogenic tissues was significantly influenced by initial tissues weight and cells density in suspension cultures; the "S" cells growth kinetics were shown during 20 days suspension culture period, and the cells viability sharply increased at first and gradually decreased in later.A better suspension culture system in masson pine was established: 1.0g tissues per 30ml fresh liquid media was the appropriate amount for the suspension culture of embryogenic cells and the mean growth rate could reach to 12.097; when cell density was 1/6 in volume in subculture, the cells showed more proliferation and better cell state; the appropriate subculture period was identified as 8-12 days. This study provided a basic information for investigating growth kinetic of suspension cultures and optimization of mass propagation system in masson pine somatic embryogenesis. It is also benefit to the genetic transformation and cell fusion in masson pine breeding improvement.
