BX3促进干细胞向神经元分化的研究

目的 探讨CBX3对小鼠胚胎干细胞的向神经元分化的作用。方法将CBX3转染小鼠ES-D3细胞后提取mRNA和蛋白,采用实时定量PCR和Westernblot的方法检测Nestin、Sox2、Gfap和TUBB3;CBX3特异性siRNA转染正在分化的小鼠胚胎干细胞(embryonic stem cells, ESCs),对照siRNA用于阴性对照,转染后提取mRNA和蛋白,采用实时定量PCR和western的方法检测Nestin、Sox2、Gfap和TUBB3。结果CBX3过表达后发现转染的ESCs干细胞特异性基因(Nestin、Sox2)mRNA及蛋白表达水平降低,而神经元分化特异性基因(Gfap、TUBB3 (tubulin beta 3 class III))mRNA及蛋白表达水平升高(P<0.05)。siRNA干扰CBX3表达后,发现相对未转染细胞,转染的ESCs的干细胞特异性基因(Nestin、Sox2)mRNA及蛋白表达水平维持高水平,而神经元分化特异性基因(Gfap、TUBB3)mRNA及蛋白表达水平较低(P<0.05)。结论CBX3有着促进胚胎干细胞分化的能力。
Objective: To investigate the effect of CBX3 on the differentiation of mouse embryonic stem cells. Methods: CBX3 was transfected into mouse ES-D3 cells, extracted mRNA and protein of ES-D3 cells, then detected the mRNA and protein level of Nestin, Sox2, Gfap and TUBB3 by real time quantitative PCR and Western-blot.And CBX3 specific siRNA was transfected into differentiated mouse ESCs. Control siRNA was used as negative control group. After transfecting into mouse ES-D3 cells, the mRNA and protein lelvel of Nestin,Sox2,Gfap and TUBB3 were detected. Results: The mRNA and protein expression of Nestin and Sox2 of transfected ESCs cells were decreased after overexpression of CBX3.But The mRNA and protein expression of Gfap and TUBB3 were increase(P<0.05). After interferencing CBX3 expression by siRNA, It was found that the mRNA and protein expression level of Nestin and Sox2 maintained a high level, but mRNA and protein expression level of the differentiation markers (Gfap and TUBB3) were low contrast to the non transfected cells (P<0.05). Conclusion: CBX3 has the ability to promote the differentiation of embryonic stem cells.
