线性聚乙烯亚胺共聚物做为潜在MR可见基因载体
Linear Poly(ethylene imine) Copolymer for Potential Magnetic Resonance Imaging and Gene Delivery
从提高基因靶向转染效率和确定靶向传输机制出发,合成了硬脂酸接枝聚乙二醇-线性聚乙烯亚胺(SA-g-PEG-lPEI),利用1HNMR、FT-IR、GPC表征聚合物,SA-g-PEG-lPEI包裹SPIO后作为潜在核磁共振显像(MRI)对比剂和基因负载的双载体, SA-g-PEG-lPEI包裹SPIO样与空白样都成囊泡状, 粒度分析表明两种聚合物自组装空白样与包裹SPIO样品粒径都在150nm左右,包裹SPIO后粒径增大;SA-g-PEG-lPEI空白样zeta-电位为+12mv左右,包裹SPIO后都有一定的降低,磁学性质表明此共聚物包裹SPIO后都仍呈现出超顺磁性,SA-g-PEG-lPEI包裹SPIO后表现出的超顺磁性和正电性使其能够作为潜在的MR可见基因载体。
In order to improve transfection efficiency and confirm target delivery mechanism, stearic acid graft Poly(ethylene glycol)-linear Poly(ethylene imine)(SA-g-PEG-PEI) copolymer was synthesized and characterized by 1HNMR, FT-IR and GPC; SA-g-PEG-PEI used as co-delivery of SPIO and gene for potential magnetic resonance imaging monitoring gene delivery, SPIO loaded with various composition of copolymers of SA-g-PEG-PEI nanoparticls(NPs) were produced using an emulsion/solvent evaporation technique, the morphology investigated by TEM of blank and SPIO-loaded SA-g-PEG-PEI NPs were vesiclar; the diameter of all the NPs were about 150nm,however, the blank NPs were smaller; the zeta-potential of blank SA-g-PEG-PEI NPs was about +12mv, but all of them decreased compared to; the SPIO-loaded NPs of SA-g-PEG-PEI exhibited superparamagetic property at room temprature. All of the results prove SPIO-loaded NPs of SA-g-PEG-PEI can act as co-delivery MRI contrast and DNA carriers, provide carriers for MRI tracking gene delivery.
