Expression of microRNA-27a-3p in peripheral blood mononuclear cells in patients with myasthenia gravis
重症肌无力(myasthenia gravis,MG)是一种由乙酰胆碱受体抗体介导的、细胞免疫依赖和补体参与的神经-肌肉接头(Neuromuscular junction ,NMJ)处传递障碍的自身免疫性疾病。研究证实,遗传因素和环境因素在重症肌无力发病中均起到重要作用,它们可共同作用打破机体自身免疫耐受状态。microRNA(miRNA)在调节免疫功能,维持免疫系统稳定方面发挥着重要作用;miRNA的异常表达可能与炎症、免疫缺陷性疫病及自身免疫性疫病密切相关。目前已有大量研究证实miR-27a-3p参与多种疾病的发生,其中以代谢性疾病、肿瘤及心血管疾病最为突出。miR-27a-3p与免疫性疾病的关系研究相对较少。本研究选择了MG患者作为研究对象,探讨miR-27a-3p与MG的相关性,为探索MG的发病机制以及诊断、治疗提供新的途径。
Myasthenia gravis (MG) are T-cell dependent antibody-mediated autoimmune disorder in which the major autoantigen is the nicotinic acetylcholine receptor (AChR) at the neuromuscular junction. Genetic and environmental factors are thought to play an important role in the pathogenesis of MG,causing tolerance breakdown through a highly complex and multifactorial process. MicroRNAs(miRNAs) have served as important regulators of various biological processes from cell proliferation and survival to organ development and immune system.Recently,studies have revealed that microRNA-27a-3p has contribute to the initiation and progression of varies of diseases, especially in metabolic disorders,tumour and CVD.Therefore,we detect the microRNA-27a-3p(miR-27a-3p) expression in peripheral blood mononuclear cells(PBMCs) of people with myasthenia gravis(MG) and the healthy control people, and to investigate the clinical significance of miR-27a-3p, in hope of illuminate our understanding of the mechanisms associated with MG presentation.?????
