叶锈菌及激素诱导对小麦叶片中Ta-SKP2A-2基因表达的影响
Expression profile of Ta-SKP2A-2 in wheat leaves infected with leaf rust pathogens and induced by hormones
为明确F-box家族基因Ta-SKP2A-2以何种信号途径参与小麦抗/感叶锈病过程。本研究在前期克隆获得Ta-SKP2A-2基因的基础上,对该基因在接种叶锈病菌及激素处理后不同时间点的表达模式进行了分析。结果表明,在TcLr38接种05-5-137③的抗病组合中,该基因被诱导表达的程度最高,接种3 h的表达量是对照的13倍;在TcLr15接种05-19-43②的抗病组合中,该基因被诱导的时间出现较晚(96 hpi),且表达量较低(为对照的2倍);在两个感病组合中,中国春(CS)中该基因的表达呈下调趋势;TcLr15与05-5-137③的感病组合中,该基因的表达呈现两个表达高峰,分别为3 hpi和96 hpi。激素处理结果表明,CS小麦中Ta-SKP2A-2基因主要受ABA的诱导上调表达,受IAA和MeJA诱导下调表达;而TcLr15小麦中该基因的表达则主要受IAA和ABA两种激素的诱导上调。结合激素处理和叶锈菌接种的结果,发现CS在叶锈菌侵染12 h前,Ta-SKP2A-2基因的总体表达呈现下调趋势,与IAA和MeJA处理的结果一致。本次试验结果在TcLr15中未发现叶锈菌接种和激素处理之间存在相同的表达模式。
In order to identify the signal pathway of F-box family Ta-SKP2A-2 genes in wheat resistance/susceptibility against leaf rust pathogen, the present study analyzed its expression profile in wheat leaves inoculated with different leaf rust strains and induced by 3 hormones. Results showed that the highest expression level of Ta-SKP2A-2 gene was observed in the combination of TcLr38 and 05-5-137③, the expression at 3h is 13 times higher than that of the control, while the induced expression is late (at 96 hpi) and lower(2 times higher than that of control) in other resistant combination of TcLr15 and 05-19-43②. In both compatible combinations, Ta-SKP2A-2 gene showed down regulation in combination of CS and 05-5-137③, and two peaks (3h and 96h) in TcLr15 inoculated with 05-5-137③. Results from hormones treatments showed that ABA can induce up-regulation of Ta-SKP2A-2, and IAA and MeJA can cause down regulation of the target gene in CS, while up-regulation of Ta-SKP2A-2 gene was mainly induced by IAA and ABA in TcLr15. Altogether, the whole tendency of Ta-SKP2A-2 gene was down regulation before 12hpi, which is same to that of IAA and MeJA. No similar expression profile was testified between inoculation with leaf rust pathogen and treatment of hormones in TcLr15.
