脑缺氧缺血增加新生大鼠海马区TLR4的表达
Brain hypoxia-ischemia increases the expression of TLR4 in hippocampus in neonatal rats
目的:观察新生大鼠缺氧缺血性脑损伤后,Toll样受体4(Toll-like receptor 4, TLR4)在海马的表达变化,为新生儿脑损伤的干预提供新的思路。方法:36只7日龄新生SD大鼠,随机分为缺氧缺血组和假手术组。缺氧缺血组结扎左侧颈总动脉,并吸入8%O2+92%N2 2小时,假手术组仅分离左侧颈总动脉,不结扎,不缺氧。两组大鼠分别于术后3d、7d、14d处死取脑(每组每时点6只),采用免疫组化、Western blot方法检测各时点海马CA1区、CA2区和CA3区TLR4的表达变化。结果:免疫组化结果显示,与对照组相比,缺氧缺血后海马CA1区、CA2区和CA3区TLR4阳性细胞数均增加,尤以CA1区增加显著,术后3d、7d两组比较差异有统计学意义(P<0.01);Western blot与免疫组化结果一致。结论:新生大鼠缺氧缺血性脑损伤后,海马区TLR4的表达增加,表明TLR4介导新生大鼠缺氧缺血脑损伤的发生。
Objective: In this study, we investigated the expression of Toll-like receptor 4 (TLR4) in the hippocampus in neonatal rat following hypoxic-ischemic brain injury. This study will provide new idea for intervention of neonatal brain injury. Methods: Thirty-six 7-day-old SD rats were divided into hypoxia-ischemia group and sham group randomly. The hypoxia-ischemia brain injury models were made by ligaturing left common carotid artery and then breathing 8%O2+92%N2 for 2 hours. The rats of sham group were only interfered with left common carotid artery dissection, without ligation and hypoxia. The brain tissues were collected at the 3d, 7d and 14d respectively. There are 6 rats in every group at each time point. The expression of TLR4 were detected by immunohistochemical staining and western blot. Results: Immunohistochemical staining showed that TLR4 positive cells were increased in hippocampal CA1 region, CA2 and CA3 region in rats with hypoxic-ischemic brain injury, compared with the control group, especially in CA1 region. There is a significant difference between the two groups at the 3d and 7d time points (P <0.01). Western blot showed the same results with immunohistochemistry. Conclusion: Expression of TLR4 is increased in hippocampus in neonatal rats following hypoxic-ischemic brain injury, which suggesting that TLR4 involved in hypoxic-ischemic brain injury.
