耐辐射奇球菌大原生质球的制备探索

Prepare large spheroplast of Deinococcus Radiodurans R1

由于大部分细菌直径比较小,研究细胞膜电生理特性的膜片钳系统不能直接对细菌的细胞进行钳制,因而细菌细胞膜中的离子通道在天然生理条件下的研究受到了阻碍。本实验采用不同抗生素对处于对数期的耐辐射奇球菌进行处理,再采用溶菌酶法将其制备成原生质球。观察形成的原生质球大小,表面光滑程度,从而筛选出作用效果比较理想的抗生素,头孢哌酮,青霉素G。在青霉素类和头孢类抗生素下,形成的原生质球的直径大其表面光滑。这类大原生质球的获得,有利于在天然生理条件下对耐辐射奇球菌细胞膜上离子通道进行研究。

he diameter of most bacterium is less than 1um. This precludes patch-clamp measurement of ion channels in bacteria directly. The research of ion channels of bacteria on the natural physiological condition is hampered. We use different antibiotics acting on the bacteria Deinococcus Radiodurans R1 which is in logarithmic phase. We get the spheroplast with lysozyme experiment. Observing the size and smooth of the spheroplast, we select several appropriate antibiotic which are useful for the large and smooth spheroplast. The large and smooth spheroplast make it possible to research ion channels of Deinococcus Radiodurans R1 on natural physiological condition.
