野生二粒小麦抗条锈病基因YrD479的定位

小麦是我国第二大粮食作物,年均播种面积约2400多万公顷,其生产却面临小麦条锈病的严重威胁。发掘和利用新型抗病基因是开展小麦抗病育种、实现种质改良的重要基础。野生二粒小麦携带丰富的抗病和抗逆基因,是小麦遗传改良的重要资源。本研究从野生二粒小麦'DIC479'中发现一个成株抗条锈病基因,暂命名为YrD479。该基因诱发侵染部位的组织坏死,有效抑制条锈菌孢子的发生。利用包含188单株的F2群体,将该基因定位在小麦2B染色体短臂,位于Xsdau018和Xsdau013之间大约4.8 cM的区段,与SSR标记Xgwm374紧密连锁,为YrD479基因的分子标记辅助选择奠定了基础。

Wheat is the No. 2 cereal crop providing staple foods in China. The annual planted area of wheat is about 24 Mha. Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici, is a devestating disease in wheat production. Breeding and growing resistant cultivars is the most economic and environmental friendly way to control the disease. Discovery and utilization of novel resistance genes is fundamental for wheat disease resistance breeding and germplasm improvement. Wild emmer wheat, Triticum dicoccoides (2n=4x=28, AABB), is a valuable genetic resource harboring untaped gene pool for disease resistance and stress tolerance. In this paper, we discovered a gene that confer adult-plant resistance to stripe rust in the wild emmer 'DIC479', and designated it as YrD479. It caused necrosis on infected tissues and prevented expansion of rust sporulation. Using a F2 population including 188 individuals, YrD479 was maped to chromosome 2BS within a 4.8 cM interval of Xsdau018 - Xsdau013, and displayed tight linkage to the SSR marker Xgwm374, which founds marker-assisted selction for the YrD479 gene.
